Ponceau S staining involves placing the membrane in the staining solution until the protein bands appear. Rinse the membrane with water after staining to remove excess dye before continuing with immunodetection.

Ponceau S is a reversible stain. The red protein bands can be washed away with water, allowing the same membrane to be used for antibody-based detection.
