Janus Green B is an oxidation-reduction indicator. In the presence of oxygen, the dye is oxidized and appears in blue, while under reduced conditions it changes to pink. This reversible property allows selective visualization of mitochondria and measurement of cellular respiratory activity.

Janus Green B stain is used for supravital staining of mitochondria, staining of nucleic acids and chromosomes, determination of cell density, and microscopic studies of various biological specimens, including fungi, yeast cells, nervous tissue, sperm cells, and tissue culture monolayers.
