Eosin B is a synthetic biological dye that is mainly used in histology, cytology and microscopy. It is used as a counterstain in combination with other stains to analyze the morphology of tissues under a microscope, which is especially useful for the study of cytoplasmic structure, connective tissues and the extracellular components of tissue.

Eosin B is water-soluble and it can also be dissolved in alcohol based solutions. It is soluble resulting in a broad range of staining applications in biological, medical and research laboratories.

Both acidic xanthene dyes, Eosin B and Eosin Y, are used in biological staining, but have different chemical structure and staining properties. Both Eosin B and Eosin Y cause a slight yellowish-pink colouration and are used routinely with H&E, with Eosin B allowing for use in some special stains.

Eosin B is safe to handle, as long as standard laboratory handling procedures are followed. But contact with the eyes, skin, or respiratory system can irritate slightly. Use appropriate personal protective equipment, such as gloves, safety glasses, etc., when handling.

Eosin B is indeed an acidic (anionic) dye. It precipitates with positively charged components of cells, like proteins in the cytoplasm, with collagen fiber, and with the connective tissues giving a typical color of pink to red during staining.
